Coomassie brilliant blue is directly proportional to protein concentration in a certain range, so it can be used for quantitative determination of protein. There are two kinds of Coomassie Brilliant Blue: G-250 and R-250. Coomassie brilliant blue G-250 is red in the free state, with the maximum light absorption at 465nm; when it combines with protein, it becomes blue, and the protein pigment combination has the maximum light absorption at 595nm. Its light absorption value is directly proportional to protein content. The binding of protein and Coomassie brilliant blue G-250 reached equilibrium in about 2 minutes, and the reaction was very rapid; the binding of protein and Coomassie brilliant blue G-250 remained stable in 1 hour at room temperature. This method was established by Bradford in 1976. The reagent is simple to prepare, easy to operate, and very sensitive. Its sensitivity is 4 times higher than Lowry method. It can be used to determine the content of microgram protein. The range of protein concentration is 0-1000 μ g / ml. it is a commonly used method for rapid determination of trace protein.
